Background
This gene is a member of the MAGEA gene family. The members of this family encode proteins with 50 to 80% sequence identity to each other. The promoters and first exons of the MAGEA genes show considerable variability, suggesting that the existence of this gene family enables the same function to be expressed under different transcriptional controls. The MAGEA genes are clustered at chromosomal location Xq28. They have been implicated in some hereditary disorders, such as dyskeratosis congenita.

Function
It is proposed to increase the ubiquitin ligase activity of the circular zinc finger E3 ubiquitin protein ligase. It can enhance the ubiquitin ligase activity of TRIM28 and stimulate p53/TP53 to be ubiquitinated by TRIM28. It is proposed to recruit and/or stabilize the ubl ligase (E2) of the E3:substrate complex. May play a role in embryonic development and tumor transformation or tumor progression. Promotes cell viability of melanoma cell lines in vitro. Autologous cytolytic T lymphocytes recognize melanoma antigens.
