Full Name Proto-oncogene c-KIT

Background The proto-oncogene c-KIT is a gene encoding a receptor tyrosine kinase protein called tyrosine protein kinase KIT, CD117 (differentiation cluster 117) or mast/stem cell growth factor receptor (SCFR).

KIT is a cytokine receptor expressed on the surface of hematopoietic stem cells and other cell types. This change in receptor form may be associated with certain types of cancer. KIT is a type III receptor tyrosine kinase that binds to stem cell factor (a substance that causes the growth of certain types of cells), also known as "steel factor" or "c-kit ligand". When the receptor binds to stem cell factor (SCF), it forms a dimer, which activates its inherent tyrosine kinase activity, which in turn phosphorylates and activates signal transduction molecules that propagate signals in the cell. After activation, the receptor is ubiquitinated to label it for transport to the lysosome and is eventually destroyed. The signal sent by KIT plays a role in cell survival, proliferation and differentiation. For example, melanocyte survival requires KIT signaling, which is also involved in hematopoiesis and gametogenesis.

Description This clone was generated using a stable CHO cell line with the FUT8 knockout gene. It was 100% non-fucosylated. In an in vitro cell-based ADCC assay, the afucosylated antibody clone showed significantly higher ADCC activity compared to its wild type version. The binding affinity of the non-fucosylated Fc variant antibody to Fc receptor is much higher than than the same antibody with fucosylated Fc. Robust and stable production of qualitative fully non-fucosylated therapeutic antibodies is achieved.
