Background
This gene encodes a kinase that activates NF-kappaB in both the Toll-like receptor (TLR) and T-cell receptor (TCR) signaling pathways. The protein is essential for most innate immune responses. Mutations in this gene result in IRAK4 deficiency and recurrent invasive pneumococcal disease. Multiple transcript variants encoding different isoforms have been found for this gene.

Function
Serine/threonine protein kinases play a key role in initiating the innate immune response against foreign pathogens. Participate in toll-like receptor (TLR) and IL-1R signaling pathway (PubMed:17878374). MYD88 is rapidly recruited into the receptor signal complex after TLR activation, and forms Myddosome with IRAK2. IRAK1 is initially phosphorylated, thereby stimulating the kinase activity and strong autophosphorylation of IRAK1. Phosphorylate the E3 ubiquitin ligase Pellino protein (PELI1, PELI2 and PELI3) to promote Pellino-mediated IRAK1 polyubiquitination. Then, the ubiquitin binding domain of IKBKG/NEMO binds to the polyubiquitinated IRAK1, which binds the IRAK1-map3k7/TAK1-TRAF6 complex and the NEMO-ikka-ikkb complex together. MAP3K7/TAK1 activates IKKs (CHUK/IKKA and IKBKB/IKKB) in turn, leading to nuclear translocation and activation of nf-kpa-b. Alternatively, TIRAP is phosphorylated to promote its ubiquitination and subsequent degradation. After LPS stimulation, NCF1 phosphorylates and regulates the activation of NADPH oxidase, suggesting a similar mechanism during microbial infection.
