Cooperation Seeking
Creative Biolabs is interested in collaborating with potential partners (include but not limit to major pharma or biotech firms) to further co-develop ADCC-enhanced Interleukin 33 antibody. For commercial partners interested in our ADCC-enhanced therapeutic antibodies, Creative Biolabs welcomes collaboration. Here are two ways for your choice, and please contact us for more details.
1) Collaborate with us and co-develop the programs from discovery phase to IND enabling. Costs will be shared.
2) Become a licensed candidate of our programs.
Looking forward to cooperating with you in the near future.
Official Name IL33
Full Name interleukin 33
Background IL33 (MIM 608678) is a member of the IL1 (see MIM 147760) family that potently drives production of T helper-2 (Th2)-associated cytokines (e.g., IL4; MIM 147780). IL33 is a ligand for IL33R (IL1RL1; MIM 601203), an IL1 family receptor that is selectively expressed on Th2 cells and mast cells (summary by Yagami et al., 2010 ).
Alternative Names IL33; interleukin 33; DVS27; IL1F11; NF-HEV; NFEHEV; C9orf26; interleukin-33; IL-33; IL-1F11; DVS27-related protein; interleukin-1 family member 11; nuclear factor for high endothelial venules; nuclear factor from high endothelial venules
Gene ID 90865
Cellular Localization Extracellular region or secreted, Nucleus
Involvement in Disease Diseases associated with IL33 include Granulomatous Mastitis and Loeffler Syndrome.
Related Pathways Its related pathways are PEDF Induced Signaling and Metabolism of proteins.
Function Cytokines bind and send signals through IL1RL1/ST2 receptors, which in turn activate the nf-kpa-b and MAPK signaling pathways in target cells. Participate in the maturation of Th2 cells and induce the secretion of T-helper type 2 related cytokines. It is also involved in the activation of mast cells, basophils, eosinophils and natural killer cells. Acts as a chemotactic agent for Th2 cells and may act as an "alert" to enhance the immune response when tissue is damaged. In the resting endothelium, the uncleaved form is expressed in large quantities, and as a chromatin-related nuclear factor, it has the characteristics of a transcription repressor. It may isolate the nuclear NF-kappaB/RELA and reduce the expression of its target protein. This form disappears quickly when angiogenesis or pro-inflammatory activation occurs.
Post-translational modifications 1.Modification sites at neXtProt 2.Modification sites at PhosphoSitePlus 3.The full-length protein can be released from cells and is able to signal via the IL1RL1/ST2 receptor. However, proteolytic processing by CSTG/cathepsin G and ELANE/neutrophil elastase produces C-terminal peptides that are more active than the unprocessed full length protein. May also be proteolytically processed by calpains. Proteolytic cleavage mediated by apoptotic caspases including CASP3 and CASP7 results in IL33 inactivation. In vitro proteolytic cleavage by CASP1 was reported but could not be confirmed in vivo suggesting that IL33 is probably not a direct substrate for that caspase.
