Background
The protein encoded by this gene is a receptor for interleukin 10. This protein is structurally related to interferon receptors. It has been shown to mediate the immunosuppressive signal of interleukin 10, and thus inhibits the synthesis of proinflammatory cytokines. This receptor is reported to promote survival of progenitor myeloid cells through the insulin receptor substrate-2/PI 3-kinase/AKT pathway. Activation of this receptor leads to tyrosine phosphorylation of JAK1 and TYK2 kinases. Two transcript variants, one protein-coding and the other not protein-coding, have been found for this gene.

Function
The cell surface receptor IL10 participates in IL10-mediated anti-inflammatory function and limits excessive tissue destruction caused by inflammation. After binding to IL10, it induces a conformational change of IL10RB, so that IL10RB also binds to IL10. The heterotetrameric assembly complex composed of the two subunits of IL10RA and IL10RB activates the kinases JAK1 and TYK2, which are related to the composition of IL10RA and IL10RB, respectively. These kinases then phosphorylate specific tyrosine residues in the intracellular region of IL10RA. Once phosphorylated, STAT3 homodimerizes and is transported to the nucleus to activate the expression of anti-inflammatory genes. In addition, il10ra-mediated STAT3 activation can inhibit starvation-induced autophagy.
