Official Name
ITGB3
Full Name
integrin, beta 3 (platelet glycoprotein IIIa, antigen CD61)
Background
The ITGB3 protein product is the integrin beta chain beta 3. Integrins are integral cell-surface proteins composed of an alpha chain and a beta chain. A given chain may combine with multiple partners resulting in different integrins. Integrin beta 3 is found along with the alpha IIb chain in platelets. Integrins are known to participate in cell adhesion as well as cell-surface mediated signalling.

Diseases associated with ITGB3 include Glanzmann Thrombasthenia and Bleeding Disorder, Platelet-Type, 16.
Related Pathways
Its related pathways are Monoamine Transport and RET signaling.
Function
Integrin alpha-v / beta-3( ITGAV:ITGB3 )It is a receptor for actin, fibronectin, laminin, matrix metalloproteinase-2, osteopontin, osteomodulin, prothrombin, platelet reactive protein, vitronectin and von Willebrand factor. Integrin alpha-iib / beta-3 (itga2b: ITGB3) is a receptor for fibronectin, fibrinogen, plasminogen, prothrombin, platelet reactive protein and vitronectin. Integrins alpha-iib / - 3 and alpha-v / - 3 recognize r-g-d sequences in a variety of ligands. The integrin alpha IIb / - 3 recognizes the h-h-l-g-g-g-a-k-q-a-g-d-v sequence in the fibrinogen chain. After activation, the integrin alpha IIb / - 3 causes platelet / platelet interaction by binding to soluble fibrinogen. This step leads to rapid platelet aggregation, which physically blocks the ruptured endothelial surface. Fibrinogen binding enhances SELP expression (through similarity) in activated platelets. ITGAV:ITGB3 It binds to fractalkine (cx3cl1) and acts as its co receptor in CX3CR1 dependent fractalkine signaling pathway. ITGAV:ITGB3 Binding to NRG1 (via EGF domain) is crucial for NRG1 ErbB Signal. ITGAV:ITGB3 Binding with FGF1, which is essential for FGF1 signal transduction. ITGAV:ITGB3 Binding with FGF2, which is essential for FGF2 signal transduction. ITGAV:ITGB3 It binds to IGF1, which is essential for IGF1 signaling. ITGAV:ITGB3 It binds to IGF2, which is essential for IGF2 signal transduction. ITGAV:ITGB3 It binds to IL1B, which is essential for IL1B signaling. ITGAV:ITGB3 The binding of pla2g2a with pla2g2a is different from the classical ligand binding site (site 1) (site 2), which induces the conformational change of integrin and enhances the binding of ligand to site 1. ITGAV:ITGB3 As a receptor for fibrillary protein 1 (FBN1), it mediates R - G - D - dependent cell adhesion to FBN1. In the brain, it plays a role in synaptic transmission and plasticity. To participate in the regulation of serotonin neurotransmission, it is necessary to localize the serotonin receptor SLC6A4 to a specific area of the synapse and reabsorb serotonin appropriately. The intensity of excitatory synapses is controlled by regulating the endocytosis of AMPAR containing Gria2, thus affecting the abundance and composition of AMPAR (through similarity). ITGAV:ITGB3 As a receptor for CD40LG.
Field of research
Cancer antibody; Cell Biology and Cellular Response antibody; Developmental Biology antibody; Immune System antibody; Signaling Transduction antibody
Post-translational modifications
Phosphorylated on tyrosine residues in response to thrombin-induced platelet aggregation. Probably involved in outside-in signaling. A peptide (AA 740-762) is capable of binding GRB2 only when both Tyr-773 and Tyr-785 are phosphorylated. Phosphorylation of Thr-779 inhibits SHC binding.

Description
This is a novel humanized single-chain antibody against a particular epitope of integrin αIIbβ3 that specifically lyses activated platelets and thus reduces the possibility of platelet-assisted metastasis. In several animal models of cancer, this antibody was able to reduce symptoms associated with metastasis. Due to the specificity of this antibody, it does not lower the platelet count significantly, nor does it induce bleeding.
