Background The product of this gene belongs to the integrin alpha chain family of proteins. Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain. This gene encodes an alpha 4 chain. Unlike other integrin alpha chains, alpha 4 neither contains an I-domain, nor undergoes disulfide-linked cleavage. Alpha 4 chain associates with either beta 1 chain or beta 7 chain.

Function Integrin alpha-4/beta-7 (Peyer patches-specific homing receptor LPAM-1) is an adhesion molecule that mediates the migration of lymphocytes and their distribution to intestinal-associated lymphoid tissue (GALT). Integrin alpha-4/beta-7 interacts with cell surface adhesion molecule MADCAM1, which is usually expressed by the endothelium of the gastrointestinal tract. It also interacts with VCAM1 and fibronectin, an extracellular matrix component. It recognizes one or more regions of the fibronectin CS-1 region. The tripeptide L-D-T in MADCAM1 interacts with L-D-V in fibronectin. Combining with HIV-1 gp120, allowing the virus to enter GALT, which is considered to be the main cause of AIDS. The interaction involves L-D-I, a tripeptide in HIV-1 gp120. The integrin alpha-E/beta-7 (HML-1) is the receptor for e-cadherin.
