Background This is a heterodimer. Its α5 subunit is 36% identical to the GPIIb subunit. This complex is located mainly on endothelial cells but also on smooth muscle cells, macrophages and platelets. Its main function is in the adhesion of cells to the extracellular matrix components.

Function Integrin alpha-5/beta-1 (ITGA5:ITGB1) is a receptor for fibronectin and fibrinogen. It recognizes the R-G-D sequence in the ligand. ITGA5: ITGB1 binds to PLA2G2A through a site (site 2) different from the classical ligand binding site (site 1), thereby inducing a change in the conformation of the integrin and enhancing the binding of the ligand to site 1. ITGA5: ITGB1 acts as a receptor for fibrillin 1 (FBN1) and mediates r-g-d-dependent cell adhesion to FBN1. ITGA5: ITGB1 is a receptor for IL1B, and its binding is essential for IL1B signaling. ITGA5: ITGB3 is the receptor for soluble CD40LG and is required for the CD40/CD40LG signaling pathway. (Microbial infection) Integrin ITGA5: ITGB1 acts as a receptor for human metapneumovirus. (Microbial infection) Integrin ITGA2: ITGB1 acts as a receptor for human parvovirus B19. (Microbial infection) In the case of HIV-1 infection, the interaction with the extracellular virus Tat protein seems to enhance the angiogenesis of Kaposi's sarcoma lesions.

In vitro, PF-5412 potently inhibited α5β1-mediated intracellular signaling, cell adhesion, migration, and endothelial cell (EC) tubulogenesis. The dual mechanism of action and robust antitumor efficacy of PF-5412 support its clinical development for the treatment of a broad spectrum of human malignancies.
