This gene encodes a tumor suppressor protein that contains a PHD-finger, which is a common motif in proteins involved in chromatin remodeling. This protein can bind TP53 and EP300/p300, a component of the histone acetyl transferase complex, suggesting its involvement in the TP53-dependent regulatory pathway. Multiple alternatively spliced transcript variants have been observed that encode distinct proteins.

The component of HBO1 complex specifically mediates the acetylation of histone H3 at the 'lys-14' site (H3K14Ac) and reduces the activity of histone H4. Through chromatin acetylation, it may play a role in DNA replication. It may inhibit tumor progression by regulating the transcriptional output of signal pathways regulating cell proliferation. When combined with rela, it inhibits the angiogenesis of brain tumor by transcriptional inhibition of rela / nfkb3 target gene. It may also specifically inhibit the loss of contact inhibition caused by activated oncogenes such as myc. Hypoxia inducible factor (HIF) activity was inhibited by interaction with HIF prolyl hydroxylase 2 (EGLN1). It can enhance the apoptosis of HC11 mammary epithelial cell line induced by serum starvation (similarity).

Citrullination by PADI4 within the nuclear localization signal disrupts the interaction with p53 and increases susceptibility to degradation.
