Description
Monoclonal antibody 12D11 (also known as clone M4d3) recognizes an epitope on the human C4dα2 chain. Complement factor C4 (molecular weight 41 kDa), formerly known as Gg protein, consists of α-, β- and γ- constituting the classical pathway of complement The activation pathway of mannose-binding lectin (MBL) converges at C4. The activated C1, MASP-1 and MASP-2 cleave C4 to form C4a and C4b, which can be cleaved by factor I. As a result, C4c and C4d are lost, all Functional sites are lost; C4d activated fragments of C4d are excellent markers for the activation of the classical complement pathway and MBL pathway. The alpha chain mediates the covalent attachment to the target surface and carries the activated form of C1 or MASP.
In addition, C4d is highly homologous to C3d, which has more than 35% shared amino acid sequence. In many diseases, such as rheumatoid arthritis (RA), hereditary angioedema (HAE), systemic lupus erythematosus (SLE) and chronic urticaria, the high complement level of C4d in serum or plasma is significantly increased. C4d levels in the plasma of patients with various humoral autoimmune diseases may also increase, and it is known that complement activation occurs. The deposition of C4d in the capillaries around the renal tubules has been shown to be a sensitive marker of antibody-mediated (humoral) rejection in renal transplantation biopsy. The monoclonal antibody 12D11 recognizes C4, C4b and C4d. The antibody 12D11 reacts with pigs, cows, baboons and rhesus monkeys.
