Monoclonal antibody 1H8 recognizes the complement factor C3b / iC3b / C3d, which specifically binds to human C3 and the breakdown products C3b, iC3b and C3dg. C3 plays a central role in the activation of the complement system. Both classical and alternative complement activation pathways require activation of it. People with C3 deficiency are susceptible to bacteria. One form of C3 convertase, also known as C4b2a, is formed by a heterodimer of C4 and C2 activated forms. It catalyzes the proteolytic cleavage of C3 produced during the activation of the classical pathway and the lectin pathway into C3a and C3b. C3a is an anaphylactic toxin, a precursor of certain cytokines (such as ASP), and C3b is a conditioning agent. Factor I can cleave C3b into C3c and C3d, the latter plays a role in enhancing the B cell response. In the alternative complement pathway, C3 is cleaved by C3bBb, which is another form of C3 convertase composed of activated forms of C3 (C3b) and factor B (Bb). Once C3 is activated to C3b, it exposes a reactive thioester, allowing the peptide to be covalently attached to any surface that can provide a nucleophile (such as a primary amine or hydroxyl). Then, activated C3 can interact with factor B. Then, factor B activates factor B to form Bb. The resulting complex C3bBb is called alternative pathway (AP) C3 convertase. C3bBb is being phased out. First, the proteolytic component of invertase Bb is removed by complement regulatory protein with decay accelerating factor (DAF) activity. Next, gradually decompose C3b into iC3b, then C3c + C3dg, and finally C3d. Factor I is a protease that uses CR1 as a cofactor for these cleavages. Clone 1H8 recognizes independent, non-overlapping epitopes on the C3 fragment. The level of C3 in the blood can be measured to support or refute a specific medical diagnosis. For example, low C3 levels are associated with certain types of kidney disease (such as glomerulonephritis and shunt nephritis after infection).

Application Notes
For flow cytometry and western blotting, the diluent to be used depends on the detection system used. It is recommended that users test reagents and determine their own optimal dilution. The typical initial working dilution is 1:50. For functional studies, the in vitro dilution must be optimized in the user's experimental setting.
