Background This gene encodes a member of the chaperonin family. The encoded mitochondrial protein may function as a signaling molecule in the innate immune system. This protein is essential for the folding and assembly of newly imported proteins in the mitochondria. This gene is adjacent to a related family member and the region between the 2 genes functions as a bidirectional promoter. Several pseudogenes have been associated with this gene. Two transcript variants encoding the same protein have been identified for this gene. Mutations associated with this gene cause autosomal recessive spastic paraplegia 13.

Function Chaperone proteins are involved in mitochondrial protein import and macromolecular assembly. Together with Hsp10, it promotes the correct folding of the imported protein. It can also prevent misfolding and correct assembly of unfolded polypeptides produced under stress conditions in the mitochondrial matrix. The functional units of these chaperone proteins are composed of seven americ loops of the large subunit Hsp60, which function as a back-to-back double loop. In the cyclic reaction, each loop of the Hsp60 loop complex binds to an unfolded substrate protein, then binds to ATP and binds to the two heptameric loops of the Hsp10 parachaperone protein. This results in the substrate protein being isolated in the lumen of Hsp60, where it can fold for a period of time without interference from other cellular components. The simultaneous hydrolysis of ATP in all Hsp60 subunits leads to the dissociation of the chaperone protein loop and the release of ADP and folding substrate proteins (possibly).
