Background Human Leukocyte Antigen (HLA) B27 (subtypes B*2701-2759) is a class I surface antigen encoded by the B locus in the major histocompatibility complex (MHC) on chromosome 6 and presents antigenic peptides (derived from self and non-self antigens) to T cells. HLA-B27 is strongly associated with ankylosing spondylitis (AS), and other associated inflammatory diseases referred to as "spondyloarthropathies".

Function The antigen presents major histocompatibility complex I (MHCI) molecules. The complex B2M/β2 microglobulin shows the α-β T cell receptor (TCR) HLA-B-restricted cd8 + T cells recognized by the main virus and tumor-derived peptide antigen presenting cells, which guide the antigen T cell immune response and eliminate infection Or transformed cells. There may also be self-peptides derived from secreted proteins or membrane protein signal sequences, although T cells directed against these peptides are usually inactivated to prevent self-reactions. Both peptides and MHC molecules can be recognized by TCR. Peptides are responsible for the good specificity of antigen recognition, and MHC residues are responsible for T cell MHC restriction. A typical intracellular peptide antigen is composed of 8-13 amino acids and is produced by intracellular proteolysis of constitutive proteasomes and immune proteasomes induced by ifgn. It can bind to different peptides containing allele-specific binding motifs, which are mainly determined by anchor residues at positions 2 and 9.
