Background The protein encoded by this gene is a homodimeric integral membrane gelatinase belonging to the serine protease family. It is selectively expressed in reactive stromal fibroblasts of epithelial cancers, granulation tissue of healing wounds, and malignant cells of bone and soft tissue sarcomas. This protein is thought to be involved in the control of fibroblast growth or epithelial-mesenchymal interactions during development, tissue repair, and epithelial carcinogenesis. Function The cell surface glycoprotein serine protease is involved in the degradation of extracellular matrix, and is involved in cellular processes such as tissue remodeling, fibrosis, wound healing, inflammation and tumor growth. Plasma membranes and soluble forms exhibited post-proline cleave endopeptidase activity, which clearly favored the Alabama/Ser-Gly-Pro-Ser/Asn Ala consensus sequence and other substrates alpha-2-antiplasmin SERPINF2 and SPRY2. It can also degrade gelatin and heat-denatured type I collagen, but cannot degrade native type I and IV collagen, vibrating protein, tendin, laminin, fibronectin, fibrin or casein. There is also dipeptidyl peptidase activity, which shows that it can hydrolyze the two residues of prolyl bonds. The n-terminal synthetic dipeptide substrate provides the penultimate residue proline, and prefers Ala-Pro, Ile-Pro, Gly-Pro, Arg-Pro Pro- Pro. The natural neuropeptide hormones used for dipeptidyl peptidase are neuropeptide Y (NPY), peptide YY (PYY), substance P (TAC1) and brain natriuretic peptide 32 (NPPB). The plasma membrane form related to DPP4, PLAUR or integrins participates in the extracellular proteolysis of extracellular matrix (ECM), and promotes cell adhesion, migration and invasion through ECM. Play a role in tissue remodeling during development and wound healing. Participate in the invasion of ECM by malignant melanoma tumor cells. Enhance tumor growth and progression by increasing angiogenesis, collagen fiber degradation and cell apoptosis, and by reducing the immune system's anti-tumor response. By degrading proteoglycan, it promotes the infiltration of glioma cells through the brain parenchyma.
