Function
Nuclear hormone receptor. Steroid hormones and their receptors participate in the regulation of eukaryotic gene expression and affect cell proliferation and differentiation in target tissues. Ligand-dependent nuclear deactivation includes direct homodimer binding to palindromic estrogen response element (ERE) sequences, or binding to other DNA transcription factors, such as AP-1/c-Jun, c-Fos, ATF-2, Sp1 and Sp3, to mediate ERE independent signal transduction. Ligand binding initiates a conformational change, allowing subsequent binding or combined binding to the multiprotein co-activator complex through the LXXLL motif of the respective components. Cross-inhibition occurs in a cell type-specific way between the estrogen receptor (ER) and nf-kpa-b. Reduce nf-kpa-b dna binding activity, inhibit nf-kpa-b-mediated transcription from IL6 promoter, and replace RELA/p65 from the promoter and related co-regulators. Recruited to the nf-kpa-b response elements of CCL2 and IL8 promoters, it can replace CREBBP. There are nf-kpa-b components RELA/p65 and NFKB1/p50 in the ERE sequence. It can also cooperate with nf-kpa-b to activate transcription related to recruiting neighboring response elements; this function involves CREBBP. Can activate the transcriptional activity of TFF1. It also mediates membrane-activated estrogen signals, involving various kinase cascades. Isoform 3 is involved in the activation of NOS3 and the production of endothelial nitric oxide. Isoforms lacking one or several functional domains are believed to regulate transcriptional activity through competitive ligand or DNA binding and/or heterodimerization with the full-length receptor. It is essential for the transcriptional regulation of BRCA1 and BCAS3 mediated by mta1. Isoform 3 can bind to ERE and inhibit Isoform 1.

The methylation may favor cytoplasmic localization. Palmitoylated (isoform 3). Not biotinylated (isoform 3). Palmitoylated by ZDHHC7 and ZDHHC21. Palmitoylation is required for plasma membrane targeting and for rapid intracellular signaling via ERK and AKT kinases and cAMP generation, but not for signaling mediated by the nuclear hormone receptor.
