Background
Ephrin receptors and their ligands, the ephrins, mediate numerous developmental processes, particularly in the nervous system. Based on their structures and sequence relationships, ephrins are divided into the ephrin-A (EFNA) class, which are anchored to the membrane by a glycosylphosphatidylinositol linkage, and the ephrin-B (EFNB) class, which are transmembrane proteins. The Eph family of receptors are divided into two groups based on the similarity of their extracellular domain sequences and their affinities for binding ephrin-A and ephrin-B ligands. Ephrin receptors make up the largest subgroup of the receptor tyrosine kinase (RTK) family. This gene encodes a receptor for ephrin-B family members.

Function
Receptor tyrosine kinases, which bind to transmembrane ephrin-B family ligands residing on neighboring cells, resulting in contact-dependent bidirectional signaling to neighboring cells. The signal pathway downstream of the receptor is called the forward signal, and the signal pathway downstream of the ephrin ligand is called the reverse signal. Generally it has overlapping and redundant functions with EPHB2. Like EPHB2, it has axon guiding functions during developmental regulation, such as the formation of corpus callosum and anterior commissural neurons, and the connection between the two main hemispheres of the temporal lobe of the cerebral cortex. In addition to its guiding role in axons, it also plays an important redundant role with other ephrin-B receptors in the development and maturation of dendritic spines and the formation of excitatory synapses. Control other aspects of development by regulating cell migration and positioning. This includes angiogenesis, palate development, and thymic epithelial development. The forward and reverse signals through the EFNB2/EPHB3 complex also regulate the migration and adhesion of cells that make the urethra tubules and divide the sulcus. Finally, it plays an important role in the differentiation of intestinal epithelial cells, separating progenitor cells from differentiated cells in crypts.
