Background
This gene is a classical cadherin from the cadherin superfamily. The encoded protein is a calcium dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. The protein functions during gastrulation and is required for establishment of left-right asymmetry. At certain central nervous system synapses, presynaptic to postsynaptic adhesion is mediated at least in part by this gene product.

Function
Calcium-dependent cell adhesion protein; it preferentially mediates the adhesion of homotypic cells by dimerizing with the CDH2 chain of another cell. Cadherin may therefore help the classification of heterogeneous cell types. As a regulator of neural stem cell quiescence, it mediates the anchoring of neural stem cells and ependymal cells in the adult subependymal zone: When MMP24 is lysed, cdh2-mediated anchoring is affected, thereby regulating the quiescence of neural stem cells. CDH2 may be involved in neural recognition mechanisms. In hippocampal neurons, dendritic spine density may be regulated.

Post-translational modifications
Cleaved by MMP24. Ectodomain cleavage leads to the generation of a soluble 90 kDa amino-terminal soluble fragment and a 45 kDa membrane-bound carboxy-terminal fragment 1 (CTF1), which is further cleaved by gamma-secretase into a 35 kDa. Cleavage in neural stem cells by MMP24 affects CDH2-mediated anchorage of neural stem cells to ependymocytes in the adult subependymal zone, leading to modulate neural stem cell quiescence (By similarity). May be phosphorylated by OBSCN.
