cadherin 1, type 1

This gene is a classical cadherin from the cadherin superfamily. The encoded protein is a calcium dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. Mutations in this gene are correlated with gastric, breast, colorectal, thyroid and ovarian cancer. Loss of function is thought to contribute to progression in cancer by increasing proliferation, invasion, and/or metastasis. The ectodomain of this protein mediates bacterial adhesion to mammalian cells and the cytoplasmic domain is required for internalization. Identified transcript variants arise from mutation at consensus splice sites.

Cadherin is a calcium-dependent cell adhesion protein. When connecting cells, they preferentially interact with themselves in an affinity manner; cadherin may therefore help the classification of heterogeneous cell types. CDH1 is involved in the regulation of cell adhesion, migration and proliferation of epithelial cells. Has a powerful invasion inhibitory effect. It is a ligand for integrin a-e / -7. E-Cad/CTF2 promotes the non-amyloid degradation of Abeta precursor. It has a strong inhibitory effect on the production of APP C99 and C83. (Microbial infection) is the receptor of Listeria monocytogenes; endogenous protein A (InlA) binds to this protein to promote the absorption of bacteria.

Acetylated. Deacetylated by SIRT2 at Lys-69 and Lys-159; deacetylation enhances the interaction of FZR1 with CDC27, leading to activation of anaphase promoting complex/cyclosome (APC/C). Phosphorylated during mitosis, probably by maturation promoting factor (MPF), leading to its dissociation of the APC/C. Following DNA damage, it is dephosphorylated by CDC14B in G2 phase, leading to its reassociation with the APC/C, and allowing an efficient G2 DNA damage checkpoint. Phosphorylated by MAK.
