Background This gene encodes activated leukocyte cell adhesion molecule (ALCAM), also known as CD166 (cluster of differentiation 166), which is a member of a subfamily of immunoglobulin receptors with five immunoglobulin-like domains (VVC2C2C2) in the extracellular domain. This protein binds to T-cell differentiation antigene CD6, and is implicated in the processes of cell adhesion and migration. Multiple alternatively spliced transcript variants encoding different isoforms have been found.

Function Cell adhesion molecules regulate the contact between heterotypic and homotypic cells by interacting with CD6. Promotes the activation and proliferation of t cells by interacting with CD6. Through the interaction with CD6, it promotes the formation and maturation of immune synapses. Mediates homotypic interaction with cells expressing ALCAM. Normal hematopoietic stem cells need to be transplanted into bone marrow. The attachment of dendritic cells to endothelial cells is mediated by homotypic interactions. It inhibits endothelial cell migration and promotes endothelial tube formation through homotypic interaction. The normal organization of the lymphatic network is required. This is necessary for the transplantation of normal hematopoietic stem cells into the bone marrow. Play a role in the hematopoietic process; normal number of hematopoietic stem cells in the bone marrow. Promote the proliferation and differentiation of osteoblasts in vitro (through similarity). Promote neurite extension, axon growth and axon guidance; axons preferentially grow on the surface containing ALCAM.
