Background
This gene encodes a member of the acid-sensing ion channel (ASIC) family of proteins, which are part of the degenerin/epithelial sodium channel (DEG/ENaC) superfamily. Members of the ASIC family are sensitive to amiloride and function in neurotransmission. The encoded proteins function in learning, pain transduction, touch sensation, and development of memory and fear. Alternatively spliced transcript variants have been described.

Function
Isoform 2 and Isoform 3 are proton-gated sodium channels; they are activated by a drop in extracellular pH and then rapidly desensitized. This channel generates a two-phase current with fast deactivation and slow continuous phase. High selectivity for sodium ions and high transport efficiency for lithium ions. Isoform 2 can also transport potassium, but it is less efficient. The larger rubidium and cesium ions are almost impenetrable. Isomer 3 can also transport calcium ions. It mediates glutamate-independent calcium (2+) into neurons during acidosis. This Ca(2+) overload is toxic to cortical neurons and may be part of the cause of ischemic brain damage. The isomer channel assembly seems to be able to adjust the channel characteristics. As a post-synaptic proton receptor, it affects the intracellular Ca(2+) concentration and calmodulin-dependent protein kinase II phosphorylation, thereby affecting the density of dendritic spines. The activity of neural circuits that regulate inner fear.
