Background
The protein encoded by this gene functions as both a phosphodiesterase, which cleaves phosphodiester bonds at the 5' end of oligonucleotides, and a phospholipase, which catalyzes production of lysophosphatidic acid (LPA) in extracellular fluids. LPA evokes growth factor-like responses including stimulation of cell proliferation and chemotaxis. This gene product stimulates the motility of tumor cells and has angiogenic properties, and its expression is upregulated in several kinds of carcinomas. The gene product is secreted and further processed to make the biologically active form. Several alternatively spliced transcript variants encoding different isoforms have been identified.

Function
Hydrolysis of lysophospholipids in the extracellular fluid produces lysophosphatidic acid (LPA) signaling molecules. The main substrate is lysophosphatidylcholine. It can also act on sphingosyl phosphorylated choline to produce sphingosyl-1-phosphate, a regulator of cell movement. It can hydrolyze pi-pnpp in vitro, pNP-TMP can be hydrolyzed to a certain extent, and ATP can barely be hydrolyzed. Involved in several motif-related processes, such as angiogenesis and neurite outgrowth. It acts as an angiogenic factor by stimulating the migration of smooth muscle cells and the formation of microtubules. Stimulates the migration of melanoma cells, possibly through a pertussis toxin-sensitive G protein. May play a role in inducing labor. May be involved in cell proliferation and fat tissue development (possibly). Tumor cell activating factor. Sn-1 lysophospholipid is cleaved to produce sn-1 lysophosphatidic acid which mainly contains 18:2 and 20:4 fatty acids. The sn-2 isomer (lyso-PAF) of 1-o-alkyl sn-glyceric acid-3-phosphocholine showed preference for sn-1.
Post-translational modifications
N-glycosylation, but not furin-cleavage, plays a critical role on secretion and on lysoPLD activity.
