Background Sclerostin is a secreted glycoprotein with a C-terminal cysteine knot-like (CTCK) domain and sequence similarity to the DAN (differential screening-selected gene aberrative in neuroblastoma) family of bone morphogenetic protein (BMP) antagonists. Loss-of-function mutations in this gene are associated with an autosomal-recessive disorder, sclerosteosis, which causes progressive bone overgrowth. A deletion downstream of this gene, which causes reduced sclerostin expression, is associated with a milder form of the disorder called van Buchem disease.

Description Romosozumab is a humanized monoclonal antibody that targets sclerostin. It is used in postmenopausal women with a high risk of fracture and in patients who have failed treatment or are intolerant to other osteoporosis treatments. Osteoporosis in the treatment of osteoporosis. Studies have shown that the drug can increase bone formation and reduce bone resorption in postmenopausal women with low bone density. Romolimumab is approved for medical use in the United States in 2019. Romosozumab is sold in the United States by Amgen under the trade name Evinity.
