The monoclonal antibody 3G11 specifically reacts with rat C6. The proteolytic cleavage of C5 by C5 convertase produces C5b, which initiates the assembly of the C5b-9 membrane attack complex. Complement component C6 is part of this C5b-9 complex. The complex is assembled from five precursor molecules in the serum. The final step in the formation of the C5b-9 complex involves the polymerization of C9, accompanied by the insertion of the complex into the cell membrane. The rat residual kidney model has been widely used to identify the mechanisms leading to the progression of kidney disease. Proteinuria is nephrotoxic and contributes to the progression of kidney damage. The activation of complement in proteinuria causes renal tubule and interstitial damage. In rats lacking C6, despite persistent proteinuria, the acute interstitial changes were resolved. This indicates that C6 mediates the chronic progression of renal tubular interstitial damage in rats with residual kidney. The monoclonal antibody 3G11 was produced by immunizing mice with C6 isolated from rat serum. The monoclonal antibody 3G11 can be used as an immunoassay to detect C5b-9 in serum and urine.
